100+ optimized IHC assays

TriStar offers immunohistochemistry (IHC) assays for oncology research on Ventana and Dako platforms, with scoring by our pathologists. Connect your assay to fit-for-purpose cohorts from our translational studies repository. We also offer IHC assay transfer and development for targets outside the library.

Assay optimization, controls and scoring

We review the assay protocol, controls and scoring criteria against your study requirements before testing begins.

Optimization
Antibody and staining conditions

Antibody clone, dilution, antigen retrieval and detection conditions are optimized using appropriate control tissue on the selected staining platform.

Controls
Controls included with each run

Positive and negative FFPE control tissue is stained alongside study samples to assess assay performance and background staining.

Validation
Validation for your intended use

Where additional validation is required, we agree the scope and acceptance criteria with you. Assessments may include specificity, sensitivity and reproducibility.

Scoring
Pathologist review and scoring

Our pathologists apply the scoring criteria agreed for your study, such as H-score or percentage of positive cells. PD-L1 scoring may use tumor proportion score (TPS) or combined positive score (CPS), as appropriate for the assay and indication.

IHC assay development for new targets

If your target is not in the library, we can assess it for assay development.

We begin by reviewing the target, tissue type and intended use of the assay. We then identify candidate antibodies and suitable positive and negative control material, including tissue from our repository where available.

Development includes antibody screening, titration, and optimization of antigen retrieval and detection on the selected automated platform. Normal tissue microarrays can be used to compare expression across nonmalignant tissues and assess staining patterns alongside tumor tissue.

We agree the scoring method and any further validation needed for your study. The final protocol and control requirements are documented before study testing.
